dc.description.abstract分子模印技術利用功能單體與模板分子的結合形成能與模板分子之形狀產生互補的辨識位置,能夠提供對目標分析物優異的特異性以及識別能力,利用此技術合成之高分子材料稱為分子模印聚合物( MIPs ) 。牛血清蛋白(Bovine Serum Albumin, BSA)是牛血清中的一種球蛋白,血液中的白蛋白主要維持滲透壓作用、pH緩衝作用、載體作用和營養作用。本篇研究的目的為開發具有高度選擇性之分子模印聚合物,針對牛血清蛋白具有高度親和力,用以從複雜的血清樣品中吸附牛血清蛋白,以達到針對血清樣品進行初步處理的效果。本研究選擇牛血清蛋白作為模板分子,並使用兩種官能基單體 MAA和 2-VP,以及交聯劑EGDMA透過總體聚合的方式合成分子模印聚合物,並利用掃描式電子顯微鏡觀察分子模印聚合物的表面特徵。之後透過對模板分子BSA的吸附能力優化合成比例及步驟。當2-VP-MIPs作為單體分子,且模板/單體/交聯劑比例在1:16:80時會有最好的吸附效果 (324 μg/mg, IF=2.12)。該MIPs具有良好的選擇性,並且能在1小時內吸附BSA,最後成功證明此MIPs材料能在多種蛋白質中選擇性去除BSA。zh_TW
dc.description.abstractThe molecular imprinting technology utilizes the combination of a functional monomer and a template molecule to form an identification position that can complement the shape of the template molecule. It can provide excellent specificity and recognition ability to the target analyte. The polymer material synthesized by the technique is called molecularly imprinted polymers (MIPs). Bovine Serum Albumin (BSA) is a globular protein in bovine serum. Albumin in the blood maintains osmotic pressure, pH buffering, carrier action and nutrition. The aim of this study was to develop a highly selective MIP with high affinity for bovine serum albumin for removing bovine serum albumin from complex serum samples. In this study, bovine serum albumin was selected as a template molecule, and two molecular monomers MAA and 2-VP, and cross-linker EGDMA were used to synthesize molecularly imprinted polymers by means of overall polymerization, and the surface characteristics of molecules were observed by scanning electron microscopy. The synthesis ratio and experimental procedures were then optimized based on the adsorption capacity of the template molecule BSA. The best result was obtained when the ratio of template/functional monomer/cross-linker was 1:16:80 using 2-VP-MIPs (324 μg/mg, IF=2.12) as monomer. This proved that the synthesized MIPs material could successfully remove abundant BSA in protein mixtures.en_US
