In this thesis, we have two topics. Firstly, we developed a spectral-domain optical coherence tomography system to obtain biological tissue scattering, the system uses coherence gating of backscattered light for tomographic imaing of tissue structure. Variation in tissue scattering due to in-homogeneities in the optical index of refraction provide imaing contrast. Finally, we have successfully used Doppler optical micro-angiography (DOMAG) to image phantom flow and skin blood perfusion in mice.
