陳頌方Chen, Sung-Fang王瀞嬴Wang, Jing-Ying2023-12-082028-08-162023-12-082023https://etds.lib.ntnu.edu.tw/thesis/detail/b13fdd8825638d45aeafad254f30946c/http://rportal.lib.ntnu.edu.tw/handle/20.500.12235/120917塑膠垃圾及汙染的議題逐漸受到重視,而其中微小的塑膠微粒可能造成的影響亦成為人們研究及關注的焦點。目前已有相關研究提出塑膠微粒可能被生物體攝入而進入循環系統並累積在各種組織中,對其產生潛在影響,甚至藉由食物鏈傳遞累積至更高的營養層級當中。本研究運用了同重元素相對與絕對定量(iTRAQ)的化學標定方法搭配液相多維分離與質譜技術,對暴露於塑膠微粒小鼠的肝臟組織進行差異蛋白質體學分析,並成功相對定量到367個蛋白質,其中在塑膠微粒投餵組中共有58個蛋白質的表現量與控制組相比具有顯著差異。根據Gene Ontology及Metascape分析,探討這些差異表達的蛋白質所參與的相關功能途徑,顯示其主要涉及細胞過程與代謝過程,以及碳水化合物代謝或能量代謝相關的途徑。這項研究可有助於進一步了解塑膠微粒的暴露對生物體的潛在風險。The issue of plastic waste and pollution has gained increasing attention, with a particular focus on the potential impacts of tiny microplastic particles. Recent research has suggested that these microplastic particles may be ingested by organisms, enter the circulatory system, and accumulate in various tissues, potentially causing adverse effects. Furthermore, there is concern regarding the potential transfer of microplastics through the food chain, leading to accumulation at higher nutrient levels. In this study, isobaric Tags for Relative and Absolute Quantitation (iTRAQ) with multi-dimensional liquid chromatography and mass spectrometry techniques were used to perform differential proteomic analysis of liver tissue in mouse exposed to microplastics. We successfully quantified 367 proteins, of which 58 proteins exhibited significant differences in expression compared to the control group in the microplastic-treated groups. Based on Gene Ontology and Metascape analysis, these differentially expressed proteins were found to be mainly involved in cellular process, metabolic process, and showed relevance to carbohydrate metabolism or energy metabolism pathways.
